Tirzepatide
Tirzepatide is a novel dual agonist of the glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP, also known as gastric inhibitory polypeptide) receptors, administered once a week. Both GLP-1 and GIP are gut-derived peptides secreted by the gastrointestinal mucosa. GLP-1 acts by binding to receptors on pancreatic beta cells to stimulate insulin secretion, thereby lowering blood glucose levels. It also delays gastric emptying and suppresses appetite, contributing to weight control. GIP, on the other hand, inhibits gastric acid and pepsin secretion, stimulates insulin release, and suppresses gastric motility and emptying, complementing the action of GLP-1 receptor agonists.
Tirzepatide combines the actions of these two insulinotropic peptides into a single molecule, representing a new class of drugs for the treatment of type 2 diabetes. It shares a similar sequence with semaglutide, with the lysine side chain modified with PEG (polyethylene glycol) to enhance water solubility and serve as a functional group for the peptide. This modification improves the stability and pharmacokinetic properties of Tirzepatide.
The primary physiological function of Tirzepatide is as a dual GLP-1 and GIP receptor agonist, designed to address the multifaceted nature of type 2 diabetes. It is currently under clinical development.
